Semax - Nootropic Neuropeptide (ACTH 4-10 Analog)
Semax is a synthetic heptapeptide derived from the ACTH (adrenocorticotropic hormone) fragment 4-10, with the addition of a Pro-Gly-Pro C-terminal tripeptide sequence that enhances metabolic stability and CNS penetration. Originally developed at the Institute of Molecular Genetics in Russia, Semax is one of the most well-characterized nootropic peptides in neuroscience research.
Mechanism of Action
Semax increases brain-derived neurotrophic factor (BDNF) and nerve growth factor (NGF) expression in the hippocampus and cortex. It modulates serotonergic and dopaminergic neurotransmission, enhances acetylcholine release, and activates MAPK/ERK signaling cascades involved in synaptic plasticity and long-term potentiation. Notably, Semax achieves its neuromodulatory activity without the adrenocortical stimulation typical of ACTH-related peptides.
